What is the principle of the Mitochondrial Apoptosis Staining Kit?
Our Mitochondrial Apoptosis Staining Kit provides a rapid two-color staining procedure performed directly with live cells. The assay quantifies the changes in mitochondrial transmembrane potential, one of the earliest intracellular events upon induction of apoptosis. It uses a cationic fluorescent dye, MitoStain, that fluoresces differently in healthy versus apoptotic cells:
- In non-apoptotic healthy cells, the dye exists as a monomer in the cytosol (green) but also accumulates and aggregates in the mitochondria, emitting a bright red fluorescence
- In apoptotic and necrotic cells, due to disruption of the mitochondrial transmembrane potential, the fluorescent dye only exists as monomer and stains the cytoplasm green.
Analysis of the stained cells can be done by FACS or fluorescence microscopy.
