Macrophage Cell Culture
The in vitro generation of monocyte-derived macrophages allows to investigate their role in numerous research applications. PromoCell provides a complete macrophage cell culture portfolio including macrophage generation media, cryopreserved human macrophages, cytokines and cell biology tools for subsequent analysis.
Assay-ready monocyte-derived human M1/M2 macrophages
Cryopreserved human macrophages are produced from human monocytes in PromoCell´s well-proven M1/M2- Macrophage Generation Media DXF and are available as non-activated, fully qualified M1- (GM-CSF) or M2- (M-CSF) polarized cells. The thawed cells plate into all tissue culture vessel formats and can be maintained as adherent, biologically functional cultures for several weeks. Optionally, user-customizable activation of the cells can be performed.
Each produced lot is tested for cell morphology, adherence rate, and viability. Furthermore, they are characterized by flow cytometric analysis of relevant markers namely CD80/CD68 for M1 macrophages (GM-CSF) and CD163/ CD68 for M2 macrophages (M-CSF).
Start in vitro generation directly from PBMCs without the need for additional CD14+ purification
The Monocyte Attachment Medium allows for the efficient adherence selection of monocytes from freshly isolated human mononuclear cells while maintaining optimal cell health. Thus, the time-consuming and costly immunomagnetic purification of monocytes is not necessary. In addition, this method offers the advantage of being selective for viable monocytes in contrast to CD14+ purification.
Full range of macrophage generation media
The PromoCell M1-Macrophage Generation Medium DXF contains GM-CSF and allows for the generation of M1-polarized macrophages, whereas the M2-Macrophage Generation Medium DXF contains M-CSF and produces M2-polarized macrophages. Both media are ready-to-use and tested in-house for successful differentiation of M1-/M2-macrophages via flow cytometry.
The Macrophage Base Medium DXF shares the same formulation as our M1/M2 Macrophage Generation Media DXF with the only exception of requiring user-added GM-CSF/M-CSF. This variant therefore offers a more cost-effective solution for users working with bulk cytokine amounts.
Defined and xeno-free (DXF) culture system
All PromoCell Macrophage Generation Media DXF are defined and xeno-free media formulations, free of all animal-derived components and substances of human origin with Human Serum Albumin as the only exception.
