Transmigration Kit
The PromoCell Invasion Kit simulates the proteolytic activity of endothelial cells and their directed migration in vitro. In this fashion, it is possible to analyze the influence of different substances on the proteolytic activity and their chemotactic efficacy in one assay.
The basis for the Kit are two compartments separated by a membrane with an accurately defined pore size of 8 μm (see Fig. 9). This membrane is covered by a matrix, which mimics the basement membrane of blood vessels. A potential chemotactic factor is placed in one compartment and a gradient develops across the membrane. Endothelial cells placed in the other compartment degrade the matrix and then migrate along this gradient. Within 24 hours after adding the test substance, the number of cells, which have migrated through the membrane, can easily be counted. This number is proportional to the chemotactic capability of the tested substance and its influence on the proteolytic activity.
The kit is ready-to-use and consists of primary endothelial cells (proliferating), coated test chambers, and optimized medium.
